"what should I do next?"
Dear Mr. Cohen,

I am desperate for your advice.
I have a very limited work experience(barely a year) and considerable educational background as a biologist. I don't have enough points(70) because I don't have any relatives in Canada nor validated job offer. Although my job search in Canada seems very promising, I have faced the difficulty of obtaining the job offer validation before.
What would be the best move for me?

With strong evidence of your employability in Canada, you may submit a skilled worker application with a request for the application of Positive Discretion. This is risky, however, and requires careful preparation.

Dear Mr. Cohen,

Thank you for the advice for my question.
Now I'd like to ask about this "positive discretion" a little more. What kind of evidences will I need to support my "employability"?
I have had a few job interviews; however, nothing has happened further because I could not obtain the job offer validation. The human resources officer's letter of the reasoning for the rejection was rather unreasonable and unfair. The incident disappointed me very much.
I hope you could give me a few suggestion.

There are no formal rules regarding the requirements of approval under "positive discretion," only that the officer must be satisfied that the points assessed under the Selection Criteria do not accurately reflect the likelihood of successfully settling in Canada.

Even in the possession of a credible written job offer, this is risky.
