1- when they check account balance
2- How to make necessary needed transactions in bank account? it means how to draw amount for purchases or expenses etc

They might ask for POF at medical and/or visa issuance, and landing.

You can do whatever you want with your account provided you can either maintain the required balance for POF or you are able to make it ready then once requested.

Yes I will be definite prove the full POF when requested.
but can I draw some amount right now. My situation is that I have started an IT business with my friend. Sometimes I will have to make payments from account for some purchases  but after some days the account is again credited after clients payments.
Due to costly purchases account goes significantly lower than required POF. but after some days it again goes above than POF amount. This rise and fall occurs usually for 15 days to 20 days.
Is it right practice till the visa issuance?

Yes no worries. CIC doesn't expect people to freeze their investments till visa issuance which usually take years.

Good luck in your IT business.

Cheers
