AINP NOC cofusion

I intend to apply under H1B category and seek help from fellow members on following questions.

Which NOC to choose?

NOC category I qualify under 2 categories i.e Biologist( moleculer biology) on the basis of my research, publications.
also as Chemist since my P.hd degree is awarded by dept of chemistry  and I have relvant documented experience as BioChemist.

What happens if my H1 expires before approval of my application( 6 year running, filing I 485 EAD etc). 

Do I need all university sealed trancripts at the time of filing with CIC?

Do I need to take IELTS having been in US for over 8 years and conducting presentations etc in english? certificate by U.S.education credential evaluator mentions clearly that medium of teaching is Engish in the universities I attended.

I could get relavant info, except for question 4 : do I need to send University sealed trancripts with CIC once I get nominated from alberta.
